Treatment Name: Phase I/II Studies of BAY 43-9006 (Sorafenib) in Combination With OSI-774 (Erlotinib), R115777 (Tipifarnib) or CCI-779 (Temsirolimus) in Patients With Recurrent Glioblastoma Multiforme or Gliosarcoma

Min Karnofsky Score: 60: Requires occasional assistance but is able to care for most of own needs
Conditions: Prior Surgery is Allowed
Prior Radiation is Allowed
Prior Chemotherapy is Allowed
Recurrent Glioblastoma Multiforme or Gliosarcoma

Comments: BAY 43-9006 (sorafenib) blocks the vascular endothelial growth factor receptor (VEGF-R) and a protein in an important tumor growth pathway (B-raf). CCI-779 (temsirolimus) blocks a specific protein, mTOR, which is important in pathways that promote tumor growth. R115777 (tipifarnib) blocks an important process called farnesylation that activates proteins for several important tumor growth pathways. OSI-774 (erlotinib) blocks the epidermal growth factor receptor (EGF-R). Combining two drugs that block these factors and receptors may provide better tumor control than a single drug. This study will also determine the relationship between biomarkers in the tumor and patient`s blood with patient response.
